Instrument Overview: MP microscopy enables optical sectioning deeper into samples thanks to the long wavelengths of excitation light. MP also allows label free imaging, meaning the auto fluorescence and the second harmonic generation (SHG; e.g. collagen fibers) of the sample can be used for 3D imaging. The inverted orientation is ideal for 3D imaging of cells/organs/tissues. The upright microscope is ideal for intravital imaging. In addition this system has a FLIM X16 bioimaging detector for fluorescence lifetime (FLIM) based applications. The measurement of fluorophore lifetime is based on Time Correlated Single Photon Counting (TCSPC) method. This enables FLIM experiments and FLIM-FRET applications. This system is equipped with both conventional PMT and highly sensitive GaAsP detectors and a Dodt microscopy detector for acquisition of transmitted light images. Both the inverted and upright microscopes are enclosed in cage-incubators to provide temperature, humidity and CO2 control.
